The research objective was to quantify at the single-molecule level the motion of fully in vitro reconstituted CMG, the eukaryotic replicative helicase. 


This was achieved by using a combination of optical tweezers and confocal fluorescence microscopy. The position along DNA molecules held in an optical trap of fluorescently labeled CMG was tracked over time, and different motion parameters were studied (e.g. diffusion constants and velocities) under different biochemical conditions.
